Expression of Cyclooxygenase-2 Protein in Basal Cell Carcinomas and Squamous Cell Carcinomas

Abstract
Cyclooxygenase(COX)-2 is one of the rate-limiting enzymes in prostaglandin synthesis. Overexpression of COX-2 has been demonstrated in various neoplasms such as colorectal cancer, lung carcinoma and esophageal carcinoma. But, the role and significance of the expression of COX-2 in non-melanoma skin cancers is not fully understood. This study aimed to determine the significance of the expression of COX-2 in basal cell carcinoma(BCC) and squamous cell carcinoma (SCC). The protein expression of COX-2 was analyzed in HaCaT cell, basal cell carcinoma and squamous cell carcinoma cell line by Western blotting. Immunohistochemical staining of COX-2 was examined on three samples of normal skin, ten samples of BCCs, and ten samples of SCCs. The immunoreactivity of COX-2 was scored from 0 to 3+, according to the intensity of the staining. Our study shows that COX-2 protein was not detected in HaCaT cell, basal cell carcinoma cell line, but was only expressed in squamous cell carcinoma cell line by Western blotting procedure. COX-2 expression was observed in 5(4: weak positive, 1: strong positive) SCC samples, but only 1(weak positive) of the 10 BCC samples and none of the 3 normal skin samples. In conclusion our results support the possibility that COX-2 might play an substantial role in the pathogenesis of human skin cancers. Further studies will be required to clarify the role of COX-2 in carcinogenesis of skin cancer.
